In the field of exosome research, the demand for accurate and reproducible methods highlights the need for first-class controls. This study aims to improve the production of Immunostep's lyophilized exosome controls, emphasising their quality as indispensable tools for biomedical research.
For exosomes manufacturing, we start from regularly genotyped cell line cultures, which guarantee the origin of the exosomes, while for exosomes from biological samples, a prior analysis against human blood-borne viruses is carried out, guaranteeing safety. The production of exosomes from cell lines is carried out in hollow fibre bioreactors, which allows better control and monitoring of CO2 and glucose in the cell culture, while a high surface-to-volume ratio and a membrane filter provide higher production and reduce contamination, respectively. In the purification process, we combine differential ultracentrifugation and size exclusion chromatography, setting a gold standard for purity in plasma/serum-derived exosomes. MISEV2018 guidelines informed our comprehensive characterisation methods, including NTA, Western Blot, Flow Cytometry and ELISA. In addition, functional studies based on cellular uptake show the therapeutic potential of labelled exosomes.
The used bottom-up cell culture method ensured authenticity, and the hollow fibre bioreactor significantly improved yield, purity and environmental control. The combination of ultracentrifugation and SEC provided exosome batches with meticulous characterisation. Quantification and global characterisation methods verified the presence of key proteins and the absence of lipoproteins, offering unprecedented insight. Functional studies highlighted the promising applications of exosomes. The freeze-drying process, using carefully selected buffers and excipients, allowed for prolonged storage without sacrificing functionality.
